  1. Product Description

SpinDesalt Column is a disposable, easy-to-operate purified product suitable for desalting, buffer replacement or small molecule removal of proteins or other large molecule samples with molecular weight greater than 5000Da.

The SpinDesalt Column contains 0.5 ml Smartdex G-25. Smartdex G-25 series medium is a kind of dextran as matrix of gel filtration chromatography medium, the main working principle is to use a mesh structure of dextran gel molecular sieves, depending on the molecular size of the separated material for separation.

  1. Matters Needing Attention

1) Remove the lower plug of the centrifugal column before use.

2) Please use 1000 g centrifuge.

3) According to the instructions, the filler is balanced using at least 2 column bed volumes of binding buffer.

4) The sample range for each column is 100-200μl. If the sample volume is less than 100μl, it is recommended that the sample be diluted with binding buffer to 100μl.

5) Note that after each step of treatment, the liquid in the pipe is sucked out in time.

6) The protein recovery rate depends on the type of protein and other biomolecules treated and is usually greater than 85%. Increasing the sample concentration can improve the recovery, and increasing the volume can also improve the recovery.

7) This product is for one use only and is not recommended for multiple uses.

 

  1. Purification Procedure

3.1 Buffer Preparation

The binding buffer can be selected as the customer. It is recommended to filter the buffers by passing them through a 0.22 μm or 0.45 μm filter before use.

3.2 Sample Preparation

The binding buffer can be selected as the customer. It is recommended to filter the buffers by passing them through a 0.22 μm or 0.45 μm filter before use.

3.3 Sample desalting

1) Remove the red plug at the bottom of the centrifugal column and put it back into the collection tube. Centrifuge 1000×g for 1 minute and pour away the protective liquid in the collection tube.

2) Put the centrifugal column back into the collection tube and add 250 μl balance liquid to the column. After all the balance liquid has penetrated into the packing, centrifuge for 1 minute at 1000×g and repeat three times.

3) Put the centrifuge column into a 1.5 mL EP tube, add 100 μl -200 μl sample, centrifuge 1000×g for 1 minute, the solution in the EP tube is the sample after desalting.
